We want to build an interior 8″ thick x 10′ long wall of cob. Should we lay a 2×4 ladder to build it on?

Do we put vapor barrier beneath the wall and above it, beneath the ceiling? In preparation we’ve already got an extra floor joist where the wall will be between the two bedrooms.

Have you calculated the weight of your 8-inch thick cob wall? I would hesitate to build a cob partition on wood framing without doing some engineering calculations. That's a heavy partition.
As I'm sure you know, cob is usually used for exterior walls rather than partitions, and usually requires a foundation of stone, concrete, or compressed earth, not wood joists.
