You want to create a seal so that the conditioned air stays within the conditioned space and the unconditioned air stays in the attic. Either works so long as you accomplish this. What I've done in the past is to use the aluminum duct tape that is used for sealing ducts (not the regular "duck" tape or off brands) and I tape the inside of the boot to the bottom of the ceiling or top of the floor so that I know the air is blowing into the conditioned space. I try to make sure any tape is obscured by the register. I'm sure there are other ways to do it but since I usually have the tape handy and it's fairly quick easy and semipermanent it seems to work out. Most houses I see have no sealant between the ceiling/floor and the boot.
